Statistical Methods for Pharmacokinetic Analyses
An Analysis of Variance (ANOVA) model will be fit to naturally logged transformed AUC0-t, Cmax and AUC0-inf with treatment, period, treatment sequence and subject nested within sequence included as fixed effects.
Separate models will be conducted only including the two treatments being compared, preserving the original period and treatment sequence.
The exponentiated least square means from each ANOVA model will be presented as the least square geometric means for each treatment.
The least square geometric mean ratios and corresponding 90% CI for AUC0-t, Cmax and AUC0-inf will also be represented graphically with all parameters on one graph.
Reference lines will indicate the 0.8000 and 1.2500 thresholds.
